RESEARCH PROFILE AND OUTSTANDING ACHIEVEMENTS
------Molecular genetics of self-incompatibility in Brassica.
1. Genomic organization of the S locus.
. I have cloned two self-incompatibility genes, SLG9 and SRK9 from an S9 homozygote of B. campestris. The two linked genes, SLG9 and SRK9, were quite similar in the long 5'-flanking regions as well as in the coding regions corresponding to the S domain, indicating that duplication or conversion may be a one of the mechanisms to produce a new set of SLG and SRK. This result has been published in "Gene" in 1997.
. I separately introduced the SLG9 and SRK9 into tobacco plants. The presence of an SLG9-like transcript in SRK9 transformants indicated the possibility of alternative polyadenylation sites in the SRK9 gene. This result has been published in "Plant and Cell Physiology" in 1996. Function of the SLG and SRK genes in the self-incompatibility recognition in stigmas was confirmed by further experiments (Takasaki et al. 2000).
. I successfully cloned the MluI-digested 76-kb fragment, containing both SLG9 and SRK9 genes, into a P1 derived artificial chromosome (PAC) vector. It is the first report of using an artificial chromosome-based vector to clone a region of the S locus. This direct cloning method is useful in analyzing the complex genomic regions. This result has been published in "Gene" in 1997.
. I dissected the 76-kb S-locus, and found 14 expressed genes in this regions. One of the genes, named SP11, encoded a small cysteine-rich protein, and was expressed specifically in anthers. SP11 was similar to a pollen coat protein, PCP-A1, which had previously been shown to bind SLG, suggesting that SP11 was a candidate of the pollen S gene. This result has been published in "Genetics" in 1999; this is the first report of the cloning of the pollen S gene in Brassica. Function of the SP11 gene in the self-incompatibility recognition in pollen was confirmed by further several experiments (Takayama et al. 2000, Watanabe et al. 2000, Shiba et al. 2001).
. I performed PFGE blot analysis to determine physical distances between the S locus genes in several S haplotypes of B. campestris and B. oleracea. The tendency for a shoter physical distance between SLG and SRK in B. campestris than B. oleracea was observed. This result has been published in "Theoretical and Applied Genetics" in 2001.

2. Molecular analysis of the S-multigene family.
. I have cloned members of the S-multigene family from an S9 homozygote of B. campestris, showing that SLG, SRK and S-related genes form a large S-multigene family in B. campestris. This result has been published in "Plant and Cell Physiology" in 1995. Furthermore, three members of the S-multigene family are clustered and linked to the S locus. The physical distance between the regions of the SLG and SRK genes and the three homologs was estimated as within 610 kb by PFGE analysis. This result has been published in "Molecular and General Genetics" in 1997. Sequence comparison and linkage analyhsis of all members of the S-multigene family were performed by further experiments (Kai et al. 2001).

------Genomic analysis in onion.
1. Constsruction of an onion BAC library.
. I constructed a bacterial artificial chromosome (BAC) libray of onion (Allium cepa). Average insert size of the library is 100 kb. A total of 48,000 clones, that correspond to 0.32 genome equivalent, were stocked in 480 glycerol stocks (corresponding to five 96-well plates), each of which contains 100 independent clones. I can screen genes of interest by PCR screening from 3D superpools. This result has been published in "Breeding Science" in 2002.

2. BAC FISH experiment in onion.
. Fluorescent in situ hybridization (FISH) screening resulted in finding the BAC clones localized on centromeric, telomeric, or several restricted chromosomal regions, although most of the clones hybridized with entire chromosomes. One of the clones showing the restricted chromosomal regions could contain 18S-5.8S-26S rDNA related sequences. The BAC FISH screening is a powerful method for identification of the molecular cytogenetic markers in large genome plants. This result has been published in "Genes and Genetic Systems" in 2001.

------Characterization of wheat centromeres.
. Cereal centromeres commonly contain many repetitive sequences that are derived from Ty3/gypsy retrotransposon. FISH analysis using a large DNA insert library of wheat identified a 67-kb clone that showed strong hybridization signals on the centromeres. The clone contains Ty3/gypsy retrotransposon-related sequences; both integrase and CCS1 family sequences were identified. Subsequently, additional 23 large-insert clones, which also contained the integrase and CCS1 sequences, were isolated. Based on the number of the integrase repeats in the clones determined by DNA gel blot analysis, I concluded that the retrotransposon-like sequences are tandemly repeated in wheat centromeres in ca. 55-kb interval on average. This conclusion is consistent with the results of FISH analysis on the extended DNA fibers. This result has been published in "Plant and Cell Physiology" in 2001.

------Genomic organization of SBEI locus in wheat.
. A wheat SBEI (Starch Branching Enzyme I) multigene family is related for the synthesis of starch. In collaboration with Dr. S. Rahman (CSIRO, Australia), I characterized the 120-kb and 100-kb clones containing SBEI genes from a large-inserts genomic library of Ae. Squarrosa. DNA gel blot analyses showed that both the 120-kb and 100-kb clones contained the same overlapped region, including SBEI-D2, -D3, and -D4. Genomic DNA gel blot analysis, PFGE analysis and Fiber FISH analysis showed that at least three SBEI-related sequences (D2,D3 and D4) were located in tandem, and another sequence similar to SBEI was possibly located near the cluster. This result has been published in "Functional & Integrative Genomics" in 2003.
